Abstract

Purpose

Monascus pigment was widely applied in food processing industry as functional additive, so more attention was paid to the fermentation optimization of pigment production. Therefore, this paper aims to evaluate the best possible fermentative conditions for maximum production of biopigment using submerged fermentation (SFM) and solid state fermentation (SSF) by Monascus purpureus HBSD 08.

Design/methodology/approach

The biopigment was produced by using an SMF and an SSF with optimized substrate to achieve higher yield. The antioxidant activity was evaluated by DPPH radical scavenging ability, superoxide anion radical scavenging ability and hydroxyl radical scavenging ability. The pigment composition was analyzed by thin layer chromatography.

Findings

Maximum Monascus pigment production (79.6 U/ml and 1,102 U/g) were obtained under an SFM and an SFF. The antioxidant activity of the pigment in an SFM was significantly higher than that in an SFM. The composition of pigment was not different in an SFM and an SFF.

Originality/value

The study developed new conditions, and Monascus strain was a candidate for producing pigment in an SFM and an SFF. To the authors’ best knowledge, this is a first attempt toward comparative evaluation on antioxidant capacity and composition between pigment in an SSF and an SFM. This result will serve for Monascus pigment production.

Abstract

Purpose

This study aims to encapsulate the crude carotene pigment isolated from waste portion of Cucurbita maxima with the help of different encapsulating agents through lyophilization to transform crude pigment into stable form for further utilization.

Design/methodology/approach

This paper opted for encapsulation of extracted carotene pigment by lyophilization using various carrier materials such as maltodextrin 20 dextrose equivalent (DE), maltodextrin 10 DE and tapioca starch along with emulsifier polysorbate-80. After encapsulation of crude carotene pigment, prepared encapsulated powder was subjected to chemical analysis. The data was analysed statistically by a complete randomized design.

Findings

Maximum encapsulation efficiency, carotene content, antioxidant activity and water solubility index were achieved when 0.06% of crude carotene pigment was emulsified with same quantity of polysorbate-80, followed by encapsulation with 20% of maltodextrin 20 DE during lyophilization.

Originality/value

Even though few researchers have worked on the encapsulation of colour pigments, no researcher has reported encapsulation of carotene pigment extracted from waste of C. maxima.

Abstract

Purpose

The purpose of this paper was to identify Serratia marcescens to extract and purify prodigiosin pigment to evaluate the antibacterial potential of the pigment.

Design/methodology/approach

Samples were collected from shrimp aquaculture ponds. Species identification was conducted using morphological, biochemical and molecular tests. Pigment extraction and purification were carried out using column chromatography. The antibacterial effect of crude and purified prodigiosin pigment was evaluated on Escherichia coli, Bacillus subtilis, Pseudomonas aeruginosa and Staphylococcus aureus as biofouling bacteria. In addition, the interaction between prodigiosin and proteins involved in biofilm formation was evaluated using molecular docking.

Findings

The results of prodigiosin extraction with solvents showed the highest percentage of pigment presence with methanol solvent in the second day of culture. The chemical structure of pure prodigiosin obtained from the column chromatography was confirmed by Fourier-transform infrared spectroscopy. Both crude and purified pigments exhibited antibacterial effects against selected bacterial strains. The antibacterial effect of the purified pigment was higher, and the highest antibacterial effect was observed on B. subtilis. Prodigiosin docking was carried out with all target proteins, and the docked energy in all of them was at an acceptable level.

Originality/value

Prodigiosin extracted from S. marcescens can be used as a bioactive compound to design and manufacture of anti-biofouling and anti-biofilm formation products to use extensively for industrial applications as a natural color in marine industries, food industry, cosmetics and textile productions.

Abstract

Purpose

Internal fluffy portion along with fibrous strands of ripe pumpkin is considered as waste in processing industries though it contains sufficient amount of ß-carotene pigment. The purpose of this paper is to use the leftover fluffy portion of ripe pumpkin (Cucurbita maxima) after the use of its flesh for the purpose of processing.

Design/methodology/approach

The data were analyzed statistically by following a complete randomized design (CRD). All analysis were performed using the software OPSTAT.

Findings

One hour pre-enzymatic treatment before solvent extraction showed significant improvement in extraction yield in comparison to the isolation of ß-carotene pigment through solvent only. Temperature time combination was optimized as 40°C for 2 h during solvent extraction to obtain maximum yield irrespective of the type of extraction method used.

Practical implications

Extracted carotene pigment can further be used as a natural food colorant in processed food products not only to enhance the color appeal but also it improves the nutritional value of the product as ß-carotene acts as a precursor of vitamin A.

Social implications

Coloring agents of natural origin are becoming famous among society due to their health benefits. Consumers are becoming reluctant to use synthetic colors because of the undesirable allergic reactions caused by them, so carotene bio-pigment produced is a natural coloring compound with wide application in the food sector.

Originality/value

Even though few researchers have worked on the extraction of carotene pigment from pumpkin, but no researcher has reported the use of a waste fluffy portion of C. maxima for extraction of ß-carotene pigment.

Abstract

Purpose

The purpose of this paper is to carry out phytochemical investigations of different extracts of Eucalyptus globulus bark such as aqueous, methanolic and supercritical carbon dioxide fluid extract (SCFE) with ethyl acetate as entrainer. Three fractions (Eu 8, 9 and 10) containing steroidal δ‐lactone were isolated from SCF extract and the structure of Eu‐10 was earlier determined on the basis of NMR, HPLC‐MS, X‐Ray crystallography.

Design/methodology/approach

Column chromatography led to the isolation of flavonoids, tannins, steroids, etc. in different solvent systems. Isolated steroidal lactone (Eu‐8,9&10) of Withanolide series were tested for the presence of total phenolic content, total flavonoid content and the results were expressed as gGAE/100 g (TPC), and gQE/100 g (TFC), respectively. The antioxidant capacity was evaluated based on their ability to scavenge free radicals generated from ABTS, DPPH, FRAP and H2O2 by spectrophotometric method.

Findings

The result of the present study showed that different extracts of E. globulus bark and the isolated fractions, exhibited different antioxidant activity. This was due to the fact that they contained different amounts of flavonoid and phenolic compounds as per their ability to solubilize these compounds; the high scavenging property of E. globulus may be attributed to hydroxyl groups existing in the phenolic compounds. All the samples exhibited different extent of antioxidant activity (AOA) and showed higher potency when compared with BHT in scavenging action of DPPH free radical. Comparative data analysis showed SCF extract to be better than methanolic and aqueous extracts, both in terms of yield and AOA, while Eu‐10 was the best amongst purified fractions.

Practical implications

The present research has serious implications on identification of natural antioxidants from E. globulus. Natural antioxidants with better structure‐activity relationship are under investigation. Isolation of withanolide from Eucalyptus bark has opened newer horizon for its use.

Social implications

Collection of Eucalyptus bark from the forest (a forest waste) by women folk can be a source of revenue generation and thus has social implication as well. It is an important agro product.

Originality/value

The steroidal lactone (Eu‐10) showed highest radical scavenging effect even at IC50, thus the isolated lactone proved to be the best potential scavenger of free radicals amongst all crude extracts and the isolated fractions.

Abstract

Purpose

The purpose of this study is to evaluate the content of bioactive pigments in coloured callus of Azadirachta indica and to understand the correlation between the callus colours with their bioactive constituents, antioxidant properties and cytotoxicity. These assessments will yield valuable insight into the use of in vitro-derived pigments for possible use as functional natural colourants.

Design/methodology/approach

In this study, the authors have successfully developed a protocol to produce leaf-derived callus of various colours with enhanced content of bioactive pigments in A. indica through plant tissue culture. Comparative analysis of the pigments content (chlorophyll, carotenoid, phenolics and anthocyanins) in the coloured callus was conducted, followed by evaluation of its bioactive properties. The antioxidant properties against 2,2-diphenyl-1-picrylhydrazyl and 2,2′-azinobis(3-ethylbenzothiazoline-6-sulfonic acid) radicals, ferric reducing antioxidant power and cytotox activities of the coloured callus extracts were also reported.

Findings

Callus of various colours were successfully produced in A. indica through plant tissue culture, and their valuable pigment content and bioactivity were evaluated. The green callus contained the highest amount of anthocyanin, followed by brown and cream callus. The total anthocyanin contents in both the green and brown callus was more than two-fold higher than that in cream callus. Contrasting observation was obtained for total phenolic content (TPC), where the TPC of cream callus was significantly higher than that in brown callus. Nevertheless, the green callus also exhibited the highest TPC. Green callus also contained the highest amount of total chlorophyll and carotenoid, as well as exhibited the highest antioxidant potential, and was found to be the only extract with active cytotox activity against SKOV-3 cells. Correlation analysis revealed that the excellent bioactivity exhibited by the coloured extracts was strongly correlated with the bioactive pigments present in the callus.

Research limitations/implications

The major bioactive compounds identified in the methanolic extracts of A. indica coloured callus are anthocyanins, phenolics, chlorophylls and carotenoids. Future research work should include improvements in the extraction and identification methods, which may lead to detection and determination of other compounds that could attribute to its bioactivity, to complement the findings of the current study.

Practical implications

This analysis provides valuable information on the application of plant tissue culture as an alternative source for sustainable production of major pigments with medicinal benefits in A. indica for possible use as functional natural colourants.

Originality/value

A comparative study on bioactive pigment production in coloured callus from A. indica leaves and its antioxidant potential and cytotoxicity is original. To the best of the authors’ knowledge, this is the first report detailing a comparative evaluation on the production of coloured callus in A. indica and its relative biochemical composition and bioactive properties.

Abstract

Purpose

This paper aims to study the effect of different organic solvents on the extraction of pigments present in callus cultures of E. cinerascens.

Design/methodology/approach

Attempts have been made to extract pigments from callus cultures through tissue culture system as an alternative replacement for conventional plant cultivation as tissue culture provides unlimited supplies of plant samples. Callus of E. cinerascens was induced from stem explant cultured in Murashige and Skoog medium supplemented with combination of 0.5 mg/L 6-benzylaminopurine and 0.5 mg/L α-naphthaleneacetic acid maintained under photoperiod of 16 h light and 8 h dark. Fresh samples of the callus were harvested and dissolved in various types and concentrations of solvents such as 100 per cent acetone, 80 per cent acetone, 95 per cent ethanol, 100 per cent methanol and 90 per cent methanol. Each of the mixtures was directly centrifuged to get clear supernatant containing pigments of interest. The pigments were detected and subsequently quantified via two simple techniques, ultraviolet-visible (UV-Vis) spectrophotometer and thin layer chromatography (TLC).

Findings

UV-Vis spectrophotometer detected two families of pigments present in the callus cultures, namely, carotenoids (carotene and xanthophyll) and tetrapyrroles (chlorophyll a and b). Pigment contents in various solvent extractions were estimated using spectroscopic quantification equations established. Through TLC, spots were seen on the plates, and Rf values of each spots were assessed to indicate the possible existence of carotenoids and tetrapyrroles.

Originality/value

This preliminary study offers significant finding for further advance research related on natural pigments extracted from E. cinerascens that would provide profits in the future applications, especially in food industry, medicine, agriculture, etc.

Abstract

The technique is presented for obtaining green pigments in the reaction of precipitation from sodium metasilicate solution and from chromium (III) salts. Sulphates and chlorides of chromium (III) as well as chromium sulphates from the tanning industry refuses were applied. The studies were directed to obtain pigments of the following composition: xCr2O3 ySiO2 zH2O. Basic conditions of precipitation process, filtration and drying of silicate pigments were established. Moreover, principal physicochemical parameters of the precipitated pigments were evaluated as well as the size and morphology of pigment particles being estimated by electron microscopy. The performed physicochemical studies aimed also at determining suitability of the pigments as fillers and carriers in oil paints.

Abstract

Purpose

In this study, it was aimed to investigate the antibacterial properties of natural pigments prepared from Thymus serpyllum.

Design/methodology/approach

Al (III), Fe (II), Sn (II) and Cu (II) complexed natural pigments were obtained by using a precipitation method and the main constituents in the pigments were identified with HPLC-DAD. Also FTIR analysis was performed for further structural characterization. Moreover, the thermal stability and thermal degradation properties of the pigments were analyzed by thermogravimetric analyses (TGA). The antimicrobial activity of the thyme plant-extracted pigments was evaluated by measuring the minimal inhibitory concentration.

Findings

Apigenin and luteolin flavones were detected as the main components of the natural dyes. Thermal degradation behaviour of the pigments was determined by means of TGA. All pigments showed high char yields and it was attributed to the high complexation between the metal and the ligand species. The antimicrobial activity of the thyme plant-extracted pigments was measured and it was found that all pigments had high antimicrobial activity. Aluminum-thymus pigments showed the highest antimicrobial efficiency among other pigments used in this study.

Originality/value

The obtained pigments have high antimicrobial activities, and therefore, they can be used for the production of antimicrobial textiles. Furthermore, Thymus-based natural pigments might have potential applications in coating, paint, plastic industries, etc.
