Abstract

Purpose

This paper aims to investigate the preventive effects of a concomitant supplementation of a lyophilized aqueous extract of Globularia alypum (Ga) leaves in a high cholesterol-diet (HC-D) on lipid profile and lecithin cholesterol acyltransferase (LCAT) activity in hypercholesterolemic rats.

Design/methodology/approach

Twenty-four male Wistar rats weighing 232 ± 10 g were divided into four groups (n = 6). Two control groups were fed a standard-diet (St-D) supplemented (C-Ga) or not (C) with 1.66% Ga leaf extract. The two others experimental groups were fed HC-D, which contains the St-D plus 1% of cholesterol and 0.5% of cholic acid supplemented (HC-Ga) or not (HC) with the same amount of Ga. At d28, feces were collected and fasting rats were anesthetized; bloods and livers were removed to measure biochemical parameters.

Findings

In hypercholesterolemic (HC) rats, Ga supplementation in HC-D induced a significant reduction in ALT (−64%, p = 0.002) and AST (−71%; p = 0.005) activities, in plasma TC (−55%; p = 0.03) and TG (−54%; p = 0.01) concentrations, in cholesterol contents of atherogenic lipoproteins VLDL (−78%; p = 0.004) and LDL-HDL1 (−64%; p = 0.003) and inversely, an increase in those of anti-atherogenic HDL2 (+14%; p = 0.002). Feeding the HC-D-Ga exhibited a reduction in atherogenic index Apo B/Apo A-I (−72%; p = 0.002), an increase in faecal lipids, cholesterol excretion and in plasma apo A-I (+60%; p = 0.002) and HDL2-cholesteryl esters (+32%, p = 0.04) and then improved LCAT activity (+31%; p = 0.03).

Originality/value

In hypercholesterolemic rats, Globularia alypum extract was effective in preventing lipid disorders by its hypolipidemic action, had an anti-atherogenic potential and a protective effect against cardiovascular risk by enhancing LCAT activity.

Purpose

The purpose of this paper is to determine the effects of isolated soy glycinin (11S) on lipid metabolism in animals subjected to a hypercholesterolemic diet.

Design/methodology/approach

Male Wistar rats were kept in individual cages under appropriate conditions. The animals were divided into three groups (n=9): normal diet (STD) given a diet containing casein as protein source, recommended in AIN‐93M; hypercholesterolemic (HC) fed a normal diet with 1 per cent cholesterol and 0.5 per cent cholic acid; and hypercholesterolemic+glycinin (HC+11S), fed a hypercholesterolemic diet, plus 11S soy protein (300 mg/kg/day), dissolved in saline and administered by gavage. After 28 days, the animals were sacrificed and blood and liver removed for biochemical analysis of total cholesterol (TC), HDL‐cholesterol (HDL‐C) and triglycerides (TG) in the plasma, hepatic TC and TG.

Findings

A single daily dose of glycinin given to the hypercholesterolemic group demonstrated its functional role, particularly in raising HDL‐C and reducing triglycerides in the liver.

Originality/value

This study demonstrates the action of the 11S globulin in soybean as a serum lipid lowering agent, in addition to its nutritional properties, especially in raising the HDL‐C.

Abstract

Purpose

This study aims to investigate the possible effect of Citrus latifolia (CL) extract on biomarkers of oxidative stress, including lipid peroxidation products in rats fed a high cholesterol diet

Design/methodology/approach

Hypercholesterolemia was induced by feeding normocholesterolemic rats 1 per cent cholesterol-enriched diet for 15 days. An experimental group (n = 20) was divided into two groups (n = 10) and fed the same diet with or without CL lyophilized aqueous extract (1 per cent) for four weeks. At day 28, ten rats from each group were killed.

Findings

Treatment with CL lyophilized aqueous extract compared with the untreated group had decreased plasma total cholesterol (TC) (−36 per cent), triacylglycerols (−48 per cent), isoprostanes values (−74 per cent) and reduced thiobarbituric acid reactive substances in erythrocytes (−21 per cent). However, the supplementation of CL peels in the hypercholesterolemic diet enhanced superoxide dismutase (+69 per cent), glutathione reductase (+30 per cent) and catalase activities (+34 per cent).

Originality/value

In hypercholesterolemic rats, administering CL extract ameliorates dyslipidemia and attenuates lipid peroxidation in tissues. These results suggest that CL could be beneficial in the primary treatment of hypercholesterolemia and oxidative damage caused by a high-cholesterol diet.

Abstract

Purpose

This paper aims to determine the effects of 11S globulin isolated from Chickpea (Cicer arietinum L.) on lipid metabolism in animals subjected to a hypercholesterolemic and hyperlipidemic diet and compared to the drug simvastatin.

Design/methodology/approach

Thirty-six male Wistar rats, kept in individual cages and under appropriate conditions, were separated into groups that were fed a normal diet (STD) containing casein as protein source and according to AIN-93G; a high-cholesterol diet (HC), normal diet plus 1 per cent cholesterol and 0.5 per cent cholic acid and 20 per cent coconut oil; HC diet plus the isolated 11S globulin (300 mg/kg/day); and HC diet plus the simvastatin (50 mg/kg/day), both dissolved in saline and administered by gavage for 28 days. After this time, the animals were killed.

Findings

The results indicated that the addition of 1 per cent cholesterol and 0.5 per cent cholic acid induced hypercholesterolemia in the animals without interfering with their weight gain. Analyses of total cholesterol (TC), HDL-cholesterol (HDL-C) and triglycerides (TG) in the plasma, and TC and TG in the liver were made. The results show that the protein isolated from chickpea, and given as a single daily dose, did not affect the levels of plasma TC and its fractions, although decreasing the TG levels. Unlike the simvastatin, the chickpea protein significantly reduced TC and TG in the liver relative to HC group.

Originality/value

A single daily dose of 11S globulin from chickpea contributed as only as additional 2.8 per cent of dietary protein intake. These findings demonstrate that 11S chickpea protein acts as a functional agent in the lipid metabolism in addition to its nutritional properties.

Abstract

Purpose

The purpose of this study was to determine the effects of chickpea (Cicer arietinum) protein hydrolysates prepared at two degrees of hydrolysis (DH) on lipoprotein profile and on oxidant status in cholesterol-fed rats.

Design/methodology/approach

Eighteen male Wistar rats (220 ± 10 g) were divided into three groups and fed for 30 days a diet containing 20 per cent casein supplemented with 1 per cent cholesterol and 0.5 per cent cholic acid. During the experimentation, the first and the second groups received daily by gavage 250 mg of chickpea protein hydrolysates/rat at DH = 8 per cent (CPH8) and DH = 17 per cent (CPH17), respectively. The third group, named control group (CG), received water under the same conditions.

Findings

Serum total cholesterol concentrations were reduced in CPH8 (p < 0.0073) and CPH17 (p < 0.0004) groups versus CG. This reduction corresponded to a lower very-low-density lipoprotein (VLDL)-cholesterol (p < 0,0019). CPH17 reduced low-density lipoprotein- and high-density lipoprotein (HDL)-cholesterol (p < 0.0001) but increased apolipoprotein A4 (p < 0.002) concentrations and lecithin-cholesterol acyltransferase activity (p < 0.0001). APOA1 remained unchanged in the treated groups. Liver total and esterified cholesterol contents were twofold lower in both treated groups versus CG. CPH8 increased triacylglycerols and phospholipids (p < 0.0001) contents, while CPH17 decreased those of unesterified cholesterol (p < 0.0016). Compared with CG, CPH8 and CPH17 reduced serum (p < 0.0001) and lipoprotein hydroperoxides by stimulating paraoxonase activity (p < 0.0001). However, only CPH17 treatment reduced serum, VLDL- and HDL-malondialdehyde contents and improved glutathione peroxidase activity (p < 0.061).

Originality/value

Thus, chickpea protein hydrolysates and especially hydrolysed at DH = 17 per cent may have a great potential for use as a nutraceutical to reduce hypercholesterolaemia and, by consequence, oxidative stress. Therefore, the degree of enzymatic hydrolysis has a significant influence on the production of potent bioactive peptides.

Abstract

Purpose

The purpose of this study is to compare the effect of olive or salmon oil on the hepatic storage and transport of fatty acids by very-low-density lipoproteins (VLDL).

Design/methodology/approach

In all, 24 male Wistar rats (80 ± 5 g) were fed a 0.5 per cent cholesterol-enriched diet with either 20 per cent casein (C) or chickpea (CP) proteins with 10 per cent olive (O) or salmon (S) oil for 28 days.

Findings

In VLDL-triacyglycerols fatty acids, oleic acid content was higher in CPS as compared to that in CS or CPO and lower in CS and CPO than that in CO; linoleic acid content was higher in all groups; arachidonic acid content was higher in CS and CPO as compared to that in CO. In the liver, TG fatty acids content was lower in CPO or CPS as compared to that in CO or CS; oleic and arachidonic acid contents were lower in CPS than that in CPO; linoleic acid content was lower in CS, CPS and CPO than that in CO, CPO and CO. In liver, phospholipid fatty acid, oleic and arachidonic acid contents were lower in CPS than that in CS; oleic, linoleic and arachidonic acid contents were lower in CPO compared to that in CO. In liver, cholesteryl esters fatty acids, oleic, linoleic and arachidonic acids contents were higher in CPS as compared to that in CS; oleic, linoleic and arachidonic acid contents were lower in CS as compared to that in CO; linoleic and arachidonic acid contents were lower in CPS than that in CPO.

Originality/value

A cholesterol-enriched diet containing casein or chickpea proteins combined with olive or salmon oil affects the hepatic storage and transport of polyunsaturated and monounsaturated fatty acids by VLDL.

Purpose

Legume seeds provide one of the most concentrated sources of nutrients rich in fibre and high‐quality protein in the diet. The present work aims to investigate the hypocholesterolemic effect of germinated Indian bean (Dolichos lablab L. var lignosus) by feeding albino rats with a hypercholesterolemic diet.

Design/methodology/approach

Male albino rats of Wistar strain were divided into five groups (each group consisting of ten rats) according to randomized block design, housed in individual cages and fed with one of the five diets over an eight‐week period. At the end of eight weeks, the body weights were recorded and the rats put down. Samples of blood and liver were collected and used for analysis of liver and plasma cholesterol.

Findings

Supplementation of the diet with dried powder of soaked bean almost brought the plasma cholesterol to 72.5 ± 0.75 from 178 ± 1.85 compared with that of the control (61.5 ± 0.70), although the liver cholesterol was still three times higher compared with the control. The 24h germinated Indian bean cotyledons could effectively counteract the effects of added cholesterol on liver and plasma by their high fibre content coupled with enormous increase in ascorbic acid levels.

Originality/value

Results suggest germination to be a simple biochemical tool to effectively lower the cholesterol levels in liver and plasma by virtue of its higher contents of ascorbic acid and dietary fibre in ischemic heart disease patients.

Abstract

Purpose

The purpose of this paper is to compare the effects of repeatedly heated coconut oil, mustard oil and sunflower oil on antioxidant status in cholesterol-fed Sprague Dawley rats.

Design/methodology/approach

The test oils were heated at 210 ± 10°C for 15 h. Male Sprague Dawley rats were divided into six groups of six animals each. In total, 15% fresh/heated oils and 1% cholesterol were mixed with the experimental diet and fed to the animals for 60 days.

Findings

Chemical analysis revealed that repeated heating of oils resulted in changes in fatty acid composition and elevated lipid peroxidation, the effects being lower in heated coconut oil. Body weight gain significantly decreased in heated coconut oil (p = 0.02), heated mustard oil (p = 0.022) and heated sunflower oil (p = 0.001) fed animals. Malondialdehyde level was significantly increased (p = 0.001) in tissues of heated oils fed animals. Concentration of protein oxidation products was significantly increased (p = 0.001) in heated oils fed animals. Activities of antioxidant enzymes were decreased (p = 0.001) in heated oils fed animals. Total thiols were decreased (p = 0.001) in tissues of animals that were fed heated oils. Animals that were fed heated mustard oil and heated sunflower oil showed lower antioxidant levels and higher oxidation products when compared to those fed heated coconut oil.

Originality/value

Studies comparing the effects of thermally oxidized oils that vary in fatty acid composition are rare. The effects of fresh and heated oils that vary in fatty acid constitution, namely, coconut oil, mustard oil and sunflower oil, in cholesterol-fed rats are studied.

Abstract

Purpose

This paper aims to study the effects of a diet supplemented with flaxseeds on dyslipidemia, oxidative stress and proinflammatory cytokines, in rats consuming a high-cholesterol diet.

Design/methodology/approach

Male Wistar rats (n = 30) weighing (250 ± 5 g) of which 10 were control and 20 were rendered hypercholesterolemic (HC) by feeding a diet enriched with 1% of cholesterol, for 15 days. After this phase, rats were divided into two groups; hypercholesterolemic group (HC) (n = 10), fed 20% casein diet enriched with 1% cholesterol; and hypercholesterolemic rats fed the same diet (n = 10), but additionally supplemented with flaxseeds (Linum usitatissimum) (Lu) powder, i.e. HC-Lu. Animals of the control group (n = 10) were fed the casein diet. All the animals were maintained on the respective diets for four weeks.

Findings

This study showed that in HC-Lu as compared to HC group, plasma total cholesterol, triacylglycerols and non-HDL cholesterol concentrations were respectively 2.4-, 1.5- and 3-fold lower. Also, the lipid peroxidation was reduced in red blood cells, organs (liver, heart and aorta) and lipoproteins (HDL2, HDL3 and VDL-LDL). A higher superoxide dismutase activity was observed in liver (+61%), heart (+62%) and aorta (+59%), whereas plasma proinflammatory cytokine (IL-1beta and IL-6) levels were decreased.

Originality/value

These results suggest that flaxseeds help to reduce hypercholesterolemia, oxidative stress and inflammation in patients with hypercholesterolemia.

Abstract

Purpose

The present investigation was undertaken to study the potential effects of milk lipids compared to sardine oil on inflammation biomarkers and lipid peroxidation in hypercholesterolemic rats. The paper aims to discuss these issues.

Design/methodology/approach

Male Wistar rats were fed 20 percent casein combined with 5 percent milk lipids or 5 percent sardine oil and 1 percent cholesterol for 28 days. A control group was fed a standard diet.

Findings

No significant difference in serum triacylglycerol (TG) was found in the milk lipids versus sardine oil and control. However, serum TG was reduced (1.7‐fold) with sardine oil compared with the control. Serum total cholesterol (TC) was, respectively, 3.6‐ and 2.5‐fold higher in milk lipids and sardine oil, respectively, compared with control. Compared to sardine oil, TC value was 1.4‐fold higher in the milk lipid. Serum C‐reactive protein (CRP) was elevated (eight‐ and 33‐fold) in the milk lipid and sardine oil compared to control, respectively. However, CRP value was four‐fold lower in milk lipids than those in sardine oil. Compared to sardine oil, iron value was two‐fold higher in milk lipids versus sardine oil. Malondialdehyde content of red blood cell, heart and brain were decreased in milk lipids versus sardine oil (p<0.05). Hydroperoxydes contents in milk lipids were also lower in heart and aorta compared to sardine oil and control (p<0.05).

Originality/value

Milk lipids compared to sardine oil does not modulate the hypercholesterolemia but decreases inflammation biomarkers and seems to protect efficiency of some tissues against the cytotoxic action and oxidative stress of cholesterol enriched diet.
