Abstract

Purpose

The purpose of this paper is to focus on the advantages of a robotic time‐lapsed microscopic imaging system for tracking stem cells in in vitro biological assays which measure stem cell activities.

Design/methodology/approach

The unique aspects of the system include robotic movement of stem cell culture flasks which enables selection of a large number of regions of interest for data collection. Numerous locations of a cell culture flask can be explored and selected for time‐lapsed analysis. The system includes an environmentally controlled chamber to maintain experimental conditions including temperature, gas levels, and humidity, such that stem cells can be tracked by visible and epifluorescence imaging over extended periods of time.

Findings

This is an extremely unique system for both individual cell tracking and cell population tracking in real‐time with high‐throughput experimental capability. In comparison to a conventional manual cell culture and assay approach, this system provides stem cell biologists with the ability to quantify numerous and unique temporal changes in stem cell populations, this drastically reduces man‐hours, consumes fewer laboratory resources and provides standardization to biological assays.

Research limitations/implications

Fundamental basic biology questions can be addressed using this approach.

Practical implications

Stem cells are often available only in small numbers – due both to their inherent low frequency in the post‐natal tissue as compared to somatic cells, and their slow growth rates. The unique capabilities of this robotic cell culture system allow for the study of cell populations which are few in number.

Originality/value

The robotic time‐lapsed imaging system is a novel approach to stem cell research.

Abstract

Purpose

Enzymes play a pivotal role in orchestrating essential biochemical processes and influencing various cellular activities in tissue. This paper aims to provide the process of enzyme diffusion within the tissue matrix and enhance the nano system performance by means of the effectiveness of enzymatic functions. The diffusion phenomena are also documented, providing chemical insights into the complex processes governing enzyme movement.

Design/methodology/approach

A computational analysis is used to develop and simulate an optimal control model using numerical algorithms, systematically regulating enzyme concentrations within the tissue scaffold.

Findings

The accompanying videographic footages offer detailed insights into the dynamic complexity of the system, enriching the reader’s understanding. This comprehensive exploration not only contributes valuable knowledge to the field but also advances computational analysis in tissue engineering and biomimetic systems. The work is linked to biomolecular structures and dynamics, offering a detailed understanding of how these elements influence enzymatic functions, ultimately bridging the gap between theoretical insights and practical implications.

Originality/value

A computational predictive model for nanozyme that describes the reaction diffusion dynamics process with enzyme catalysts is yet not available in existing literature.

Abstract

Purpose

Institutional theorists treat law and regulations as external factors that is part of the organization’s environment. While institutional theory has been criticized for its inability to recognize the role of agents and to theorize agency, the growing literature on institutional work and institutional entrepreneurship, partially informed by and co-produced with practice theory, advances a more dynamic view of processes of institutionalization. In order to cope with legal and regulatory frameworks, constituting the legal environment of the organization, there are evidence of organizational responses in the form of bargaining, political negotiations, and decoupling of organizational units and processes. The purpose of this paper is to report how legal and regulatory frameworks both shape clinical practices while at the same time they are also informed by the activities and interests of professional communities and commercial clinics.

Design/methodology/approach

This paper reports an empirical study of the Swedish-assisted conception industry and is based on a case study methodology including the use of interviews and formal documents and reports issues by governmental agencies.

Findings

The empirical material demonstrates how scientists in reproductive medicine and clinicians regard the legal and regulatory framework as what ensures and reinforces the quality of the therapies. At the same time, they actively engage to modify the legal and regulatory framework in the case when they believe it would benefit the patients. The data reported presents one successful case of how PGD/PGS can be used to develop the efficacy of the therapy, and one unsuccessful case of regulatory change in the case of patient interest groups advocating a legalization of commercial gestational surrogacy. In the former case, scientific know-how and medicinal benefits served to “push” the new clinical practice, while in the latter case, the “demand-pull” of patient interest groups fails to get recognition in regulatory and policy-making quarters.

Originality/value

The study contributes to the literature on agency in institutional theory (e.g. the emerging literature on institutional work) by emphasizing how legal and regulatory frameworks are in a constant process of being modified and negotiated in the face of novel technoscientific practices and social demands. More specifically, this process include many scientific, technological, economic, political and social relations and resources, making the legal environment of organizations what is the outcome from joint negotiations and agreements across organizational and professional boundaries.

Abstract

Anxiety, depression and impaired health-related quality of life (HRQoL) are commonly reported in patients with multiple sclerosis (MS) and are of great interest for therapeutic approaches. Based on regional differences a quantitative assessment of these factors in comparison to the general population, and the consideration of demographic cofactors, would be useful when designing specific interventions. We adopted such an approach in a German cohort of MS patients. Anxiety, depression (HADS) and HRQoL (SF-36) were measured in 49 consecutive outpatients with MS and compared to age- and gender-adjusted control groups (n=1330 for HADS; n=5087 for SF-36) extracted from German National Health Surveys. Patients with MS showed significantly increased levels of anxiety and depression as well as decreased HRQoL with the exception of mental health; the effect sizes ranged from 0.39 (depression) to 1.06 (physical functioning). As could be expected, MS patients with relapsing-remitting clinical course had better physical functioning than patients with secondary progressive MS. There were strong relations between anxiety and depression (r=0.54; P<0.01), and between neurological impairment (EDSS) and physical functioning (r=-0.80; P<0.001) as well as depression (r=0.48; P<0.05). This investigation of MS patients confirms the prevalence and impact of anxiety, depression and most of the HRQoL dimensions in MS patients and provides evidence for the usefulness of a quantitative comparison to a region-specific general population as a starting point for therapeutic approaches.

Abstract

Purpose

This paper aims to develop a novel micro-ablation system to realise micrometric and well-defined hydrogel structures. To engineer a tissue it is necessary to evaluate several aspects, such as cell-cell and cell-substrate interactions, its micro-architecture and mechanical stimuli that act on it. For this reason, it is important to fabricate a substrate which presents a microtopology similar to natural tissue and has chemical and mechanical properties able to promote cell functions. In this paper, well-defined hydrogel structures embedding cells were microfabricated using a purposely developed technique, micro-laser ablation, based on a thulium laser. Its working parameters (laser power emission, stepper motor velocity) were optimised to produce shaded “serpentine” pattern on a hydrogel film.

Design/methodology/approach

In this study, initially, swelling/contraction tests on agarose and alginate hydrogel in different solutions of main components of cell culture medium were performed and were compared with the MECpH model. This comparison matched with good approximation experimental measurements. Once known how hydrogel changed its topology, microstructures with a well-defined topology were realised using a purposely developed micro-laser ablation system design. S5Y5 neuroblastoma cell lines were embedded in hydrogel matrix and the whole structure was ablated with a laser microfabrication system. The cells did not show damages due to mechanical stress present in the hydrogel matrix and to thermal increase induced by the laser beam.

Findings

The hydrogel structure is able to reproduce extracellular matrix. Initially, the hydrogel swelling/contraction in different solutions, containing the main components of the most common cell culture media, was analysed. This analysis is important to evaluate if cell culture environment could alter microtopology of realised structures. Then, the same topology was realised on hydrogel film embedding neuronal cells and the cells did not show damages due to mechanical stress present in the hydrogel matrix and to thermal increase induced by the laser beam. The interesting obtained results could be useful to realise well-defined microfabricated hydrogel structures embedding cells to guide tissue formation

Originality/value

The originality of this paper is the design and realisation of a 3D microfabrication system able to microfabricate hydrogel matrix embedding cells without inducing cell damage. The ease of use of this system and its potential modularity render this system a novel potential device for application in tissue engineering and regenerative medicine area.

Abstract

Purpose

Exposure to a public health threat of significant proportions made current models inadequate to explain the failure phenomenon in small businesses. Hence, the need to reimagine the phenomenon. Borrowing from the principles of biology, this study extended theoretical and empirical perspectives on the failure phenomenon by unpacking its constituent elements and the measurement metrics using the regeneration lens.

Design/methodology/approach

Based on a cohort tracked over time, the study estimated the survival probabilities of small and medium-scale enterprises (SMEs) with and without regeneration using the Kaplan–Meier method. The study investigated the factors that predict enterprise regenerative capacity using the multivariate Cox proportional hazard ratios.

Findings

Rates of interruption in business activity, by month, ranged between 0% and 18% during the follow-up period. True mortality rates hovered between 0% and 4% over the same period. Over three in five SMEs that experienced interruption in business activity without ceasing operations regenerated at some point in time during the follow-up period. The survival probabilities beyond the follow-up period were 0.85 and 0.44 with and without regeneration effects, respectively. Fresh capital injection (+), the introduction of new/improved processes or products/services (+), perceived business outlook (+) and the presence of debt (−) influenced the capacity to regenerate.

Research limitations/implications

The cohort was followed for only six months. There is a need to continue interrogating the failure phenomenon in other contexts over longer periods using the regeneration lens. Bringing on board academia, financial institutions and other SME-related ecosystem players will be strategic.

Practical implications

The approach provides a more nuanced understanding of the life and well-being of enterprises under conditions of disruption. Improving the precision and validity of failure-related statistics enhances their utility in policy and remediation-related discussions.

Social implications

The results did not show significant differences in SME mortality rates between male and female-owned enterprises. The results provide further evidence that the failure phenomenon is ungendered. As such, financial institutions and the SME ecosystem at large must eliminate perceptual gender biases in the financing and other support to SMEs.

Originality/value

The study used the principles of biology to reimagine the failure phenomenon in small businesses. The approach breathes life into entrepreneurship research and policy.

Abstract

This chapter is an ethnographic exploration of birthing and body politics in the United States and Uganda with the placenta as the catalyst for understanding reproductive regulation and gendered bodily epistemologies. Based on fieldwork spanning 2009–2017 with rural, traditional midwives in Southern Uganda, merged with recent, anecdotal observations from Los Angeles County and greater California and the United States generally, this work considers cultural terrains of placentas as well as corresponding worldviews and perspectives, ranging from life-generating organ imbued with vast spiritual and physiological significance, to preventative mental health food, to bio-waste that is incinerated or filled with toxic chemicals. The bio-ontologies of placentas are explored herein in terms of toxic contingencies and with regard to the relationship between health and industry.

Toxic entanglements and embodied politics of risk and exposure explored herein point to dehumanizing and ill-fitting regulations that stifle health autonomy and medical sovereignty. Such disempowering governance is compounded by gender and myriad cultural factors. With implications for national and international policies, this work examines my findings that illustrate ways in which flesh, technologies and knowledge intersect in bio-praxes that monitor and manage, rather than support, the reproductive body. This work suggests departure from colonial instability and dispossession by re-scripting medicine in such a way that achieves health justice through bodily knowledge, or enfleshed understandings. Decolonizing the flesh demands ungripping health encounters from praxes of control, in favour of choice and preference. This entails reclaiming physiologies as well as reimagining how medical systems inform core ethos.
