Abstract

Purpose

Increased ethanol accumulation during ethanol fermentation generates stress in yeast cells, which finally reduces the fermentation performance and efficiency. Trehalose, a potential stress protectant, has been reported to regulate the response of yeast to diverse environmental stresses. This study aimed to explore how exogenous trehalose application affects the survival, transcriptome and antioxidant enzymes of Wickerhamomyces anomalus grown under ethanol stress conditions.

Design/methodology/approach

Exogenous trehalose was applied to the growth condition of W. anomalus, and optical densitometric method was used to detect contents of intracellular trehalose and MDA and activities of CAT and SOD. The survival was evaluated using spot analysis. Differentially expressed genes (DEGs) were identified through transcriptomics analysis.

Findings

The results showed that ethanol stress induced the accumulation of intracellular trehalose, with further exogenous trehalose application improving the survival and alleviating oxidative stress in ethanol-stressed W. anomalus. Transcriptomic results showed that trehalose has pleiotropic regulating effects on ethanol-stressed W. anomalus since most DEGs annotated to energy metabolism, amino acid metabolism, translation, folding, sorting and transport were affected post trehalose addition. Therefore, it is found that trehalose protected W. anomalus against ethanol stress, and these findings provide interesting insights into the mechanistic role of trehalose in improving ethanol stress tolerance of W. anomalus.

Originality/value

(1) Protective effect of exogenous trehalose addition on the survival of ethanol-stressed W. anomalus was proved. (2) Exogenous trehalose addition could partly alleviate oxidative stress induced by ethanol stress and affect transcriptome in W. anomalus.

Abstract

Purpose

Nations such as the USA are investing in technologies such as electronic health records in order to collect, store and transfer information across boundaries of health care, public health and research. Health information brokers such as health care providers, public health departments and university researchers function as “access points” to manage relationships between the public and the health system. The relationship between the public and health information brokers is influenced by trust; and this relationship may predict the trust that the public has in the health system as a whole, which has implications for public trust in the system, and consequently, legitimacy of involved institutions, under circumstances of health information data sharing in the future. This paper aims to discuss these issues.

Design/methodology/approach

In this study, the authors aimed to examine characteristics of trustors (i.e. the public) that predict trust in health information brokers; and further, to identify the factors that influence trust in brokers that also predict system trust. The authors developed a survey that was administered to US respondents in 2014 using GfK’s nationally representative sample, with a final sample of 1,011 participants and conducted ordinary least squares regression for data analyses.

Findings

Results suggest that health care providers are the most trusted information brokers of those examined. Beliefs about medical deceptive behavior were negatively associated with trust in each of the information brokers examined; however, psychosocial factors were significantly associated with trust in brokers, suggesting that individual attitudes and beliefs are influential on trust in brokers. Positive views of information sharing and the expectation of benefits of information sharing for health outcomes and health care quality are associated with system trust.

Originality/value

This study suggests that demonstrating the benefits and value of information sharing could be beneficial for building public trust in the health system; however, trust in brokers of information are variable across the public; that is, knowledge, attitudes and beliefs are associated with the level of trust different individuals have in various health information brokers – suggesting that the need for a personalized approach to building trust.

Abstract

Purpose

Gliomas are common intracranial tumors with the characteristic of diffuse and invasive growth. The prognosis is poor, and the recurrence rate and mortality are higher. With the development of big data technology, many methods such as natural language processing, computer vision and image processing have been deeply applied in the medical field. This can help clinicians to provide personalized and precise diagnosis and therapeutic schedule for patients with different type of gliomas to achieve the best therapeutic effect. The purpose of this paper is to summarize and extract useful information from published research results by conducting a secondary analysis of the literature.

Design/methodology/approach

The PubMed and China National Knowledge Infrastructure (CNKI) literature database were used to retrieve published Chinese and English research papers about human gliomas. Comprehensive analysis was applied to conduct this research. The factors affecting survival and prognosis were screened and analyzed respectively in this paper, and different methods for multidimensional data of patients were discussed.

Findings

This paper identified biomarkers and therapeutic modalities associated with prognosis for different grade of gliomas. This paper investigated the relationship among these clinical prognostic factors and different histopathologic tying and grade of gliomas by comprehensive analysis. This paper summarizes the research progress of biomarker in medical imaging and genomics of gliomas to improve prognosis and the current status of treatment in China.

Originality/value

Combined with multimodal data such as genomics data, medical image data and clinical information data, this paper comprehensively analyzed the prognostic factors of glioma and provided guidance and evidence for rational treatment planning and improvement of clinical treatment prognosis.

Purpose

Mixed fermentation with Saccharomyces cerevisiae and non-Saccharomyces yeasts has become an oenlogical tool to improve wines’ organoleptic properties. However, the maximum utilization of this method is dependent upon understanding the influence of mixed cultures on the physiology of S.cerevisiae and non-Saccharomyces yeasts.

Design/methodology/approach

In this study, the supernatants from 48 h mixed-culture fermentation were added to the pure cultures of Issatchenkia orientalis and Saccharomyces, respectively. And the authors used RNA sequencing to determine the transcriptome change of I.orientalis and S.cerevisiae in a mixed culture.

Findings

The results showed that multiple genes associated with cell growth and death were differentially expressed. Genes related to biosynthesis of amino acids were enriched among those upregulated in the mixed-fermentation supernatant. Meanwhile, the differential expression level of genes encoding enzymes essential for formation of aroma compounds was found in the single and in the mixed fermentation. The high expression level of molecular chaperones Hsp70, Hsp90 and Hsp110 suggests that metabolites of mixed-culture fermentation may lead to aggregation of misfolded proteins. Moreover, upregulation of ethanol dehydrogenase I ADH1 in the mixed-culture fermentations was highlighted.

Originality/value

This is the first time that RNA-seq was used to analyze changes in the transcriptome of mixed cultures. According to the results the authors’ manuscript provided, an integrated view into the adaptive responses of S.cerevisiae and non-Saccharomyces yeasts to the mixed-culture fermentation is benefit for the potential application of S.cerevisiae and non-Saccharomyces yeasts in fruit wine brewing.

Abstract

Purpose

This paper aims to supply an introduction to the bioinformatics discipline for information professionals, outlining how current information management issues are hampering the effective integration and interoperability of resources.

Design/methodology/approach

The approach taken is to outline some of the more challenging issues to illustrate their consequences, such as syntactic and semantic heterogeneity, data storage formats and media, and the existence of inconsistencies in information content in bioinformatics resources. A discussion of these topics indicates how semantic web concepts and technologies, together with e‐science initiatives, can be used to address these problems.

Findings

The paper reveals that, if one considers the use of semantic web technologies such as XML and ontologies for the development of information standards that allows integration of different information systems, these systems could then be placed into applications such as web services and GRIDS tailored for biological studies. Such applications would provide automated functionality for database integration, workflow management, inclusion of provenance data, and notification of services.

Originality/value

The value of this paper is that it exemplifies how information professionals can make an impact on the discipline of bioinformatics, which historically has not been addressed by experts with information skills.

Abstract

Purpose

The purpose of this article is to evaluate the development and use of the gene ontology (GO), a scientific vocabulary widely used in molecular biology databases, with particular reference to the relation between the theoretical basis of the GO, and the pragmatics of its application.

Design/methodology/approach

The study uses a combination of bibliometric analysis, content analysis and discourse analysis. These analyses focus on details of the ways in which the terms of the ontology are amended and deleted, and in which they are applied by users.

Findings

Although the GO is explicitly based on an objective realist epistemology, a considerable extent of subjectivity and social factors are evident in its development and use. It is concluded that bio-ontologies could beneficially be extended to be pluralist, while remaining objective, taking a view of concepts closer to that of more traditional controlled vocabularies.

Originality/value

This is one of very few studies which evaluate the development of a formal ontology in relation to its conceptual foundations, and the first to consider the GO in this way.

Abstract

Purpose

This study aims to identify the current barriers to sustainability in the bioscience laboratory setting and to determine which mechanisms are likely to increase sustainable behaviours in this specialised environment.

Design/methodology/approach

The study gathers qualitative data from a sample of laboratory researchers presently conducting experimentation in the biological sciences. A questionnaire, regarding sustainability in the laboratory, was developed and distributed to all bioscience researchers at Aberystwyth University.

Findings

Although the majority of respondents had favourable attitudes to sustainability, almost three‐quarters (71 per cent) stated that they were not conducting their research in the most sustainable way possible. The factors most likely to hinder sustainable behaviour were lack of support, lack of information and time constraints. However, monetary costs and benefits, closely followed by “other” costs and benefits, were most likely to encourage sustainable behaviour in the laboratory.

Research limitations/implications

There is a need to extend the present research to other types of biological research, such as field‐based studies. Different biological disciplines may have different consumable requirements and waste streams, thereby changing the barriers to sustainability observed.

Practical implications

The findings have immediate practical implication for higher education institutions wishing to adopt researcher‐approved mechanisms to reduce the environmental impact of biological laboratory research.

Originality/value

This is the first study to design a sustainability questionnaire which is specific to research scientists and laboratory users. The paper is therefore of immense value to the numerous global higher education institutions with working laboratories which seek to minimise the environmental impact of research.

Abstract

Purpose

This study aims to understand how Big Data foster a greater acceptance of performance management systems (PMS) discourses in health care.

Design/methodology/approach

This paper focusses on the case of head and neck cancer treatment and prevention and benefits from the analysis of archival sources and 19 interviews with physicians in the field. It uses the framework of the Middle Range theory (MRT) to understand whether, in the case of head and neck cancer, Big Data may favour the enactment of PMS discourses in health care, in turn benefiting from any improvement in PMS.

Findings

This study setting unveils the changing pathway known as reorientation through boundary management. Medical professionals internalized and even mobilized PMS discourses, showing the premises for evolutionary changes in the future, when the current limitations will be dealt with.

Originality/value

This paper offers new theoretical, practical and policymaking insights into how new technologies can foster positive PMS discourses among actors who usually resist them. This value also extends to different fields and contexts.

Abstract

Purpose

The purpose of this paper is to explore the perceptions and experiences of early adopters of the technology.

Design/methodology/approach

Registered dietitians (RDs) (n=14) were recruited from the UK, Canada, South Africa, Australia, Mexico and Israel. Six qualitative interviews and two focus groups were conducted online using a conference calling platform. Data were recorded, transcribed and thematically analyzed.

Findings

Early adopters of nutrigenomics (NGx) were experienced, self-efficacious RDs who actively sought knowledge of NGx through communication with one another and the broader scientific community. They considered NGx an extension of current practice and believed RDs had the skills to deliver it. Perceived barriers to widening the application of NGx were linked to skepticism among the wider dietetics community. Proliferation of unregulated websites offering tests and diets was considered “pseudoscience” and detrimental to dietetics fully embracing NGx. Lack of a sustainable public health model for the delivery of NGx was also perceived to hinder progress. Results are discussed with reference to “diffusion of innovation theory.”

Originality/value

The views of RDs who practice NGx have not been previously studied. These data highlight requirements for future dietetic training provision and more inclusive service delivery models. Regulation of NGx services and formal recognition by professional bodies is needed to address the research/practice translation gap. Further research is required to inquire as to the views of the wider dietetics profession.
