Abstract

Purpose

The purpose of this paper is to evaluate the antioxidant activity and toxicity of Clinacanthus nutans leaves from three conventional extraction methods, i.e. maceration, Soxhlet and magnetic stirring.

Design/methodology/approach

Total flavonoid content (TFC) and phenolic content (TPC) were determined using colorimetric method of aluminum chloride and Folin-Ciocalteu (FC) assay, respectively. Antioxidant property of C. nutans was evaluated using 2,2'-diphenyl-1-pierylhydrazyl (DPPH) free radical scavenging assay. Cytotoxic activity of C. nutans against brine shrimp was evaluated based on LC50 (lethality concentration) after 24 h exposure to the plant extract.

Findings

The highest TPC of C. nutans was observed with Soxhlet extraction method (98.87 ± 10.43 mg of gallic acid equivalents (GAE/g) followed by maceration (68.77 ± 2.45 mg of GAE/g) and magnetic stirring (46.75 ± 2.45 mg of GAE/g). Interestingly, remarkable highest TFC was observed with magnetic stirring (568.90 ± 4.85 mg of rutin equivalent (RE)/g) followed by maceration (249.60 ± 2.79 mg of RE/g) and Soxhlet (174.8 ± 1.74 mg of RE/g). On the other hands, the extract obtained using maceration method showed the highest antioxidant activity (IC50: 14.18 mg/mL compared to ascorbic acid 144.36 µg/mL). Cytotoxicity of C. nutans from all extraction methods showed similar LC50 values with maceration (3.81 mg/mL), Soxhlet (2.61 mg/mL) and magnetic stirring (4.56 mg/mL), respectively.

Originality/value

Both phenolic and flavonoids are responsible for the antioxidant activity, of C. nutans extracts. Based on Meyer's toxicity index, all extracts were nontoxic (LC50>1 mg/mL).

Purpose

Kefir is a probiotic grown with milk, with a slightly sour flavor and has been consumed for thousands of years. Kefir grains contain bacteria and yeast. In the past, kefir was administrated as a drug against tuberculosis, cancer and gastrointestinal disorders. The purpose of this paper is to investigate the potential anticancer properties of kefir and its ability to affect natural killer cells' (NKCs') activity.

Design/methodology/approach

The assay of cytotoxic activity of NKCs by cytometric analysis was used, which included four stages: isolation of natural killers; quantification of target cells; incubation of natural killers with target cells at ratios of 12.5:1, 25:1 and 50:1 in CO2 incubator; and measurement of cells with flow cytometer. The same procedures were repeated, but the third stage was modified with the addition and incubation of 50, 75, and 100 μL kefir (of 24 hour culture with 3.5 per cent fresh milk) with K562 and leiomyosarcoma cells lines, and kefir and NK cells with K562 or LMS cell lines.

Findings

The results showed that kefir's cytotoxic activity without the presence of NKCs reached an average of 85 per cent in both cell lines. With the addition of NK cells in kefir, the cytotoxic activity further increased by 10 per cent. Kefir alone did not cause any statistically significant death in NK cells. Kefir seems to have significant cytotoxic action by itself without stimulating NK cells in a significant manner. However, further studies are needed to establish the role of kefir in the prevention and treatment of neoplasmatic diseases.

Originality/value

The paper provides information and new data, for nutritionists and clinical dietitians, about the effects of kefir in the prevention of cancer.

Abstract

Purpose

Whey is a by-product of paneer, cheese and casein industry and considered as a dairy waste. Worldwide, approximately 180–1,900 million tons of whey is produced annually. Whey is classified as a high pollutant due to its organic matter level. Owing to its high chemical oxygen demand and biological oxygen demand, it is a big threat to the environment. Whey contains 4.5%–5.0% lactose, 0.6%–0.7% protein, 0.4%–0.5% lipids, vitamins and minerals. Due to its high nutritional profile, it is a good substrate for the microorganisms for production of natural pigments. The purpose of this paper is to review the utilization of low cost substrate (whey) for production of various types of pigments and their applications in different sectors.

Design/methodology/approach

The databases for the search included: Scopus, PubMed, Science Direct, Web of Science, Research gate and Google. The main search was directed towards different types of natural pigments, stability, technologies for enhancing their production and contribution towards circular economy. Approximately 100 research papers were initially screened. A global search was conducted about natural pigments. Research articles, review papers, books, articles in press and book chapters were the type of search for writing this review paper.

Findings

Production of natural pigments using whey and their addition in food products not only improves the colour of food but also enhances the antioxidant properties of food products, helping the health benefits by chelating free radicals from the body. The sustainable use of whey for production of natural pigments can improve the bio-based economy of different industries and thereof the national economy.

Originality/value

Efficient utilization of whey can bring a lot more opportunities for production of natural pigments in a sustainable manner. The sustainable approach and circular economy concepts will benefit the dependent industries and health conscious consumers. The potential uses of whey for the production of natural pigments using diverse organisms are highlighted in this paper.

Abstract

Purpose

This study aimed to evaluate the cytotoxic/apoptotic effects of sweet apricot kernel ethanolic extract (SAEE) on human cancerous PANC-1 and 293/KDR normal cells.

Design/methodology/approach

The extract was prepared by maceration, and its chemical composition was analyzed by gas chromatography-mass spectrometry (GC-MS). The biological effects of SAEE on PANC-1 and 293/KDR cells were investigated using MTT (3–(4, 5-dimethylthiazole-2-yl)-2,5-diphenyl tetrazolium bromide) assay, DAPI (4',6-diamidino-2-phenylindole) and AnnexinV/propidium iodide (PI) staining. The expression of pro- and anti-apoptotic genes was evaluated by real-time quantitative polymerase chain reaction (real-time q-PCR) analysis.

Findings

The SAEE showed the selective growth inhibitory activity against PANC-1 cells with an IC50 (the 50% inhibitory concentration) value of about 1 mg/mL at 72 h. Further investigations by DAPI staining and flow cytometry revealed nucleus fragmentation and elevation of apoptotic cells, respectively. Also, a significant decrease in B-cell lymphoma 2 (Bcl-2)/Bcl-2-associated x protein (Bax) ratio (0.41, p = 0.001) and the up-regulation of caspase-3 expression (1.5 fold, p = 0.002) indicated the induction of apoptosis in PANC-1 cells but not in 293/KDR non-cancerous cells. These results suggest that SAEE could induce apoptosis in cancer cells via a mitochondrial dependent pathway. Furthermore, GC-MS analysis showed that the SAEE is rich in γ-sitosterol and γ-tocopherol. Overall, the findings suggest that because of the selective impacts of SAEE on PANC-1 cells, it can be considered as a supportive care in adjuvant therapy for pancreatic cancer. However, the potent anticancer effects of main components of SAEE and its clinical value as an antitumor drug should be further investigated.

Research limitations/implications

Considerable limitations of this study were that the related mechanisms of selective impacts of SAEE on cancerous and normal cells and potent cytotoxic/apoptotic effects of γ-sitosterol and γ-tocopherol as major components of SAEE were not investigated.

Originality/value

Recently, a growing interest has been dedicated to plant-based natural products. Sweet apricot kernel exerts a number of pharmacological activities; however, the anticancer effect, related mechanisms and its active compounds were rarely investigated. In this study, the authors aimed to evaluate the cytotoxic/apoptotic effects of SAEE on human cancerous PANC-1 and 293/KDR normal cells.

Abstract

Purpose

Research on cancer chemopreventive properties of fruits has increased in recent years. Polyphenols have been suggested to exert such effects. The purpose of this paper is to determine the cytotoxic activity of Mangifera pajang (bambangan) and Artocarpus odoratissimus (tarap) crude extracts against selected cancer cell lines (i.e. ovarian, liver and colon cancer) and to compare the amount of selected polyphenols (phenolic acids, flavanones, flavonols and flavones) in the kernel, peel and flesh of M. pajang; and the seed and flesh of A. odoratissimus.

Design/methodology/approach

Cytotoxicity activity of the extracts are investigated using MTT (3‐(4,5‐dimethylthiazol‐2‐yl)‐2,5‐diphenyltetrazolium bromide) assay while polyphenols are determined using high performance liquid chromatography.

Findings

The results show that only the kernel and peel extracts from M. pajang display cytotoxic activity in liver and ovarian cancer cell lines with IC50 values ranging from 34.5 to 92.0  μg/ml. The proliferation of colon cancer cell line is inhibited only by the kernel of M. pajang with IC50 value of 63.0  μg/ml. The kernel and peel from M. pajang contains a broad range of polyphenol phytochemicals which might be responsible for the cytotoxicity activity against selected cancer cell lines.

Originality/value

Previous reports have indicated that both M. pajang and A. odoratissimus contain high antioxidant properties. This study further determines the phytochemicals profiling in both fruits, which might contribute to the antioxidant activity. Besides that, the result from this study shows that the waste of the fruits (i.e. kernel and peel) contain superior phenolic phytochemicals and display better anticancer potential compared to the flesh; suggests the use of them in health‐industry application. Utilization of all parts of the fruits (i.e. flesh, seed, kernel and peel) for the development of nutraceutical and functional food application is suggested.

Abstract

Purpose

The purpose of this paper is to screen cytotoxic activities of commonly used culinary plants in Malaysia, Pandanus amaryllifolius (daun pandan), Curcuma longa (turmeric leaves) and Etlingera elatior (kantan flower) against selected cancer cell lines.

Design/methodology/approach

Plant samples were extracted exhaustively with ethanol and concentrated under rotary evaporator. Cytotoxic evaluation was carried out with plant extracts (0‐100 μg/ml) using 72‐h MTT assay.

Findings

Exposure of plant extracts reduced cell viability of HepG2 (hepatocellular carcinoma), HT‐29 (colon carcinoma), MDA‐MB‐231 (non‐hormone‐dependent breast cancer), MCF‐7 (hormone‐dependent breast cancer) and HeLa (cervical cancer); 50 percent inhibitory values (IC50) were obtained for MDA‐MB‐231, HepG2, HT‐29. Extracts within the concentrations of 10‐100 μg/ml were found not to be effective against proliferation of MCF‐7 and HeLa.

Originality/value

The paper shows how culinary plants – daun pandan, turmeric leaves and kantan flower displayed selective cytotoxic effects towards liver, colon and breast cancer in vitro.

Abstract

Purpose

The study aimed to disclose the anti-cancer activity of Pluchea indica tea leaves by evaluating the cytotoxicity on breast and cervical cancer cells, compared with non-cancer cells.

Design/methodology/approach

Two P. indica extracts were prepared using two solvents, namely hot water (PA) and ethanol (PE). MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide) and clonogenic assays were applied to determine cytotoxic effect of both extracts toward cancer cells from human breast (MDA-MB-231 and MCF7) and cervix (SiHa, HeLa and C-33A) and also non-cancer Vero cells. Dichlorofluorescein diacetate (DCFDA)-staining assay was used to quantify the intracellular level of the reactive oxygen species (ROS). Correlation between the quantity of compounds present and the cytotoxicity of the extracts was analyzed by Pearson's method and a possible class of bioactive compounds was proposed based on the highest correlation coefficient (r).

Findings

Significant reduction in cell viability and proliferation capability was observed in all cancer cells after treatment with either PA or PE extract albeit PE was more effective. Lower toxicity was detected in Vero cells, indicating the selectivity and safety of extracts. The intracellular ROS level was augmented in treated cancer cells which were inversely correlated to cell viability, suggesting the cancer toxicity was likely induced by intracellular oxidative stress. As flavonoids were found abundantly in the extracts and flavonoids' content was the most related to the activity (r = 0.815), it was hypothesized that the flavonoids might play crucial roles in cancer cytotoxicity.

Originality/value

P. indica tea-leaf extracts can be a good source of promising anti-cancer agents with reduced side effects for breast and cervical cancer treatment.

Abstract

Purpose

The purpose of this paper is to summarize the scientific information of various qualities of bael fruit juice used in traditional system of medicine for variety of purposes. Utilization of bael fruit juice in day-to-day life has great nutritional, therapeutic, and commercial importance. Bael fruit contains nutrients like vitamins (riboflavin), minerals, trace elements, energy and phytochemicals, including flavonoids, polyphenols and antioxidants, that have been shown to have varied health benefits. In past few decades, bael has been extensively studied for its medicinal properties by advanced scientific techniques, and a variety of bioactive compounds like marmelosin, tannins, alkaloids, coumarins, steroids, rutacine, y-sitosterol, psoralin, xanthotoxin, scopolotein, aegelemine, aegeline, marmeline, fragrine, dictamine, cinnamide and different derivatives of cinnamide have been isolated from its fruit juice.

Design/methodology/approach

The medicinal value of bael fruit is very high when the harvests just begin to ripen. As a result, it has a high demand as alternative medicine for curing the diseases like diabetes, high cholesterol, peptic ulcer, inflammation, diarrhea and dysentery, constipation, respiratory infection. Furthermore, the bael fruit juice has anticancer, cardio protective, antibacterial, antifungal, radio protective, antipyretic, analgesic, antioxidant, antiviral, anthelmintic and anti-inflammatory, hepatoprotective, wound healing properties. The ripe fruit juice is aromatic, has cooling and laxative effects, and arrests secretion or bleeding.

Findings

The unripe or half-ripe fruit juice is good for digestion, useful in preventing or curing scurvy, and it strengthens the stomach action. It helps in the healing of ulcerated intestinal surfaces and has appreciable activity against intestinal pathogenic organisms. The present review summarizes the scientific information of various qualities of bael fruit juice used in traditional system of medicine for a variety of purposes.

Originality/value

It is quite evident from this review that bael is an important medicinal herb and extensively used in Ayurveda, Siddha and other medicinal systems. Bael fruit juice is an excellent source of water and natural sugar and is important principally for containing vitamins, minerals, phytochemicals, antioxidants, pigments, energy, organic acids, dietary fiber and other food components, which are the key factors in the medicinal value of this plant. Moreover, mechanisms of action of a few bioactive compounds have been identified so far.

This study aims to focus on the possibility of developing new thiazole azo dyes with good colouristic application properties, biological and pharmacological activities.

Design/methodology/approach

Coupling of curcumin with different aromatic diazonium salts of 2-amino thiazole derivatives, such as 2-aminobenzothiazole, 2-amino-5-phenylthiazole, 2-amino-5-methylthiazole and 2-amino-5-nitrothiazole-produced novel azo dyes. Structures of all synthesised dyes were fully confirmed via spectroscopic and analytical methods. Those compounds were examined for their antimicrobial, anticancer and antioxidant activities. They were applied on polyester fabrics and, subsequently, their dyeing properties, light, washing, perspiration, rubbing and sublimation fastness were determined.

Findings

Prepared dyestuffs were suitable for dyeing polyester fabrics. It was found that all prepared dyes possess high colour strength, as well as good overall fastness properties. Meanwhile, the synthesised compounds exhibited good biological and pharmacology activity.

Research limitations/implications

Synthesis of these four azo dyes for textile dyeing was not conveyed earlier.

Practical implications

Thaizolyl disperse dyes were responsible for giving better colour assessment and fastness properties on polyester fabrics.

Social implications

Although, most of synthesis eco-friendly dyes are expensive, they are showing a good antimicrobial and anticancer activity.

Originality/value

It gave straightforward approach to synthesise novel thiazolyl azo dyes with good biological, pharmacology activities, good colour assessment, and fastness properties.

Abstract

Purpose

The present study aims to focus on the possibility of developing new eco-friendly azo dyes with good colouristic application properties, exhibiting biological and pharmacological activities.

Design/methodology/approach

Coupling of 4-hydroxycoumarin with a variety of aromatic diazonium salts of 2-aminothiazole, 2-aminobenzothiazole, 4-aminoantipyrine, 4-aminoacetophenone, adenine sulphate, a-naphthylamine and sulphadimidine to produce novel azo dyes. The compounds were fully characterised using spectroscopic and analytical methods. All of the compounds were tested for their antimicrobial, anticancer and antioxidant activities. The prepared dyestuffs were dyed on polyester fabrics and subsequently their dyeing properties, light, washing, perspiration, rubbing and sublimation fastness were determined.

Findings

The spectroscopic data of the synthesised compounds have provided decisive evidence that such compounds exist in the solid state as the azo-dike to form C and in solution in equilibrium tautomer forms A, B and D. The prepared dyestuffs are suitable for either heat transfer printing or traditional printing on polyester and nylon 6 fabrics. The prints obtained from the dyes possess high colour strength, as well as good overall fastness properties. Also the synthesised compounds exhibit good biological and pharmacology activity.

Research limitations/implications

Synthesis of these seven azo dyes for textile dyeing had never been reported previously.

Practical implications

The dyestuffs derived from 4-hydroxycoumarin are reasonable azo disperse dyestuffs giving good all round fastness properties on polyester fabrics.

Social implications

Production of less expensive and new eco-friendly dyes exhibit antimicrobial and anticancer activity.

Originality/value

It provided a potentially simple way to synthesize novel coumarin azo-dyes exhibit good biological and pharmacology activity and also exhibit good overall fastness properties.
