Abstract

Purpose

The purpose of this study is to evaluate the effects of flaxseed (L. usitatissimum [Linn]) on blood pressure, redox status markers and acetylcholinesterase enzyme activity in ouabain-induced hypertension in normal Wistar rats.

Design/methodology/approach

Male Wistar rats weighing 250 ± 7 g (n = 24) fed with an experimental diet containing 20 per cent casein were divided into three groups (n = 8) and received a daily subcutaneous injection of either 0.9 per cent saline solution (T group) or 10 µg/kg/day of ouabain diluted in saline solution-treated Oub-Lu or not with 1 per cent of flaxseed (L. usitatissimum) mixed in the diet for 21 days.

Findings

The results showed that treatment with flaxseed had a significant effect (p < 0.05) in decreasing systolic, diastolic blood pressure, mean arterial pressure and the heart rate in hypertensive rats. Total and non-HDL cholesterol levels were reduced by –26 and –35 per cent; p = 0.04, respectively. Concentrations of thiobarbituric acid-reactive substances were significantly decreased by –85 and –42 per cent (p = 0.001 and p = 0.04), respectively in the plasma and heart. Nitric oxide levels were increased in the aorta (+ 63 per cent; p = 0.001). Moreover, in the heart and aorta, a significant increase was noted in the thiol contents (+81 and +69 per cent; p = 0.001, respectively), glutathione peroxidase (+50 per cent; p = 0.03 and p = 0.01, respectively) and acetylcholinesterase activities (75 and +19 per cent, respectively; p = 0.001 and p = 0.04).

Originality/value

These results suggest hypotensive, cardiomoderating and antioxidant effects of flaxseed in ouabain-induced hypertension in the rat. In addition, it promotes a significant increase of the acetylcholinesterase activity in tissues.

Abstract

Purpose

Because of the bioaccumulation effect, organophosphorus pesticides cause long-term damage to mammals, even at small concentrations. The ability to perturb the phospholipid bilayer structure as well as the overstimulation of cholinergic receptors makes them hazardous to humans. Therefore, there is a need for a quick and inexpensive detection of organophosphorus pesticides for agricultural and household use. As organophosphorus pesticides are acetylcholinesterase (AChE) inhibitors, biosensors using this mechanism hold a great promise to meet these requirements with a fraction of reagents and time used for measurement comparing to laboratory methods. This study aims to manufacture AChE-coated, screen-printed carbon electrodes applicable in such amperometric biosensors.

Design/methodology/approach

AChE enzyme, known for catalytic activity for the hydrolysis of acetylthiocholine (ATCh), could be used to obtain electrochemically active thiocholine from acetylthiocholine chloride in aqueous solutions. Using Malathion’s inhibitory effect towards AChE, pesticides’ presence can be detected by reduction of anodic oxidation peaks of thiocholine in cyclic voltammetry.

Findings

The conducted research proved that it is possible to detect pesticides using low-cost, simple-to-manufacture screen-printed graphite (GR) electrodes with an enzymatic (AChE) coating. Investigated electrodes displayed significant catalytic activity to the hydrolysis of ATCh. Owing to inhibition effect of the enzyme, amperometric response of the samples decreased in pesticide-spiked solution, allowing determination of organophosphorus pesticides.

Originality/value

Printed electronics has grown significantly in recent years as well as research focused on carbon-based nanocomposites. Yet, the utilization of carbon nanocomposites in screen-printed electronics is still considered a novelty in the market. Biosensors have proved useful not only in laboratory conditions but also in home applications, as glucometers are a superior solution for glucose determination for personal use. Although pesticides could be detected accurately using chromatography, spectroscopy, spectrometry or spectrophotometry, the market lacks low-cost, disposable solutions for pesticide detection applicable for household use. With biosensing techniques and electric paths screen-printed with GR or graphene nanocomposites, this preliminary research focuses on meeting these needs.

Abstract

Purpose

The purpose of this paper is to analyse how novel homicide defences predicated on contemporary neuroscience align with legal insanity.

Design/methodology/approach

Doctrinal analysis, systematic investigation of relevant statutes and cases, was used to elucidate how the law of insanity is evolving. Cases represent the first recorded instance of a particular neuroscientific defence. US appellate cases were categorised according to the mechanism of action of neurotransmitter relied upon in court. A case study approach was also used to provide a contextualised understanding of the case outcome in depth.

Findings

Findings broadly depict how the employment of expert testimony runs parallel with our contemporary understanding of key neurotransmitters and their function in human behaviour. Generally, medico-legal evidence concerning neuromodulating agents and violent behaviour was inconclusive. However, the outcome of defence strategy may depend on the underlying neurotransmitter involved.

Practical implications

This study shows that as more discoveries are made about the neurobiological underpinnings of human behaviour; this new knowledge will continue to seep into the US court system as innovative defence strategies with varying success. Medical and legal practitioners may gauge the success of a defence depending on the neuromodulating agent.

Originality/value

Many scholars have focused on the role of neuroimaging as neuroscientific evidence and how it is used is shaping US criminal jurisprudence. To the best of the author’s knowledge, no study has incorporated the true origin of neuroscientific evidence as being underpinned by the understanding of neurotransmitters.

Abstract

Purpose

Mushrooms have always been treasured and appreciated, thanks to their nutritional as well as medicinal properties. The purpose of this paper is to reveal these properties such as antioxidant activities and enzyme inhibition potentials of three mushroom species from Ordu, Turkey. Furthermore, phenolic profiles which are believed to be source of these properties are investigated.

Design/methodology/approach

The assays followed to determine total phenolic, flavonoid and antioxidant contents, antioxidative and enzyme inhibition activities are spectrophotometric methods. Phenolic profiles of mushroom samples were investigated by using the HPLC system.

Findings

The highest phenolic content was found for Lactarius deliciosus. Pyrogallol was the major phenolics in all three samples. Total phenolic content values supported that finding. But, total flavonoid content was the maximum for Cantharellus cibarius. The magnitude of the antioxidant activities revealed by the different tests varied between species. Furthermore, inhibition potential degrees on acetylcholinesterase, butrylcholinesterase, tyrosinase and urease activities were worth mentioning. Lactarius pyrogalus had anti-tyrosinase activity with a 26.229 percent inhibition rate. IC50 values for urease inhibition effect of mushroom samples were in the range of 0.37–0.90 mg/mL.

Originality/value

Enzyme inhibitory potentials for L. pyrogalus was revealed first time. On the other hand, studies on anti-tyrosinase activity of mushrooms are very rare, while almost no studies have been conducted on anti-urease activity. So, the results of present study are very valuable. Thus, the obtained data not only increase the ecological importance of the region but also provide significant contributions to the literature in terms of new sources rich in antioxidants.

Purpose – Description of the effects of the revolution in neuroscience and other areas of biology that can help to explain the roots of some portion of violent crime. The chapter reconsiders the role of brain chemistry in social behavior and violent behavior. To illustrate the interdisciplinary complexities entailed when linking brain chemistry to policy decisions concerning violent crime, this analysis has four main stages: first, why might SiFs (H2SiF6 and Na2SiF6, jointly called “silicofluorides” or SiFs) be dangerous? Second, what biochemical effects of SiF could have toxic consequences for humans? Third, on this basis a research hypothesis predicts children in communities using SiF should have increased uptake of lead from environmental sources and higher rates of behavioral dysfunctions known to be caused by lead neurotoxicity.

Design/methodology/approach – To illustrate the implications of the new issues involved, this chapter focuses on a public policy that inadvertently seems to increase rates of violent crime. Since violent behavior is one of the effects of lead neurotoxicity, the hypothesis is tested using multiple sources of data including rates of violent crime studied using a variety of multivariate statistical techniques (including analysis of variance, multiple regression, and stepwise regression).

Findings – Various data sources point to greater violence among individuals with greater exposure to SiFs.

Originality/value – Testing hypotheses linking neurotoxins to violent behavior reveals the generally unsuspected value of analyzing human social behavior and public policy from the perspective of evolutionary psychology.

Abstract

Sheep dip toxicity from organophosphate is a problem affecting at least 40 people, and probably far more in the farming community in north Devon. It is a problem not simply of acute exposure, but also of sub‐acute and possibly chronic exposure, with significant neurological and other illness. The epidemiology of this exposure and the pathological characteristics need to be elucidated. The toxicology of organophosphate chemicals in man, in low dosage, needs further investigation. Presently, farmers should not use organophosphates if they have had symptoms from previous exposure. To enable there to be no financial penalty, governments should ensure that there is no price differential between organophosphate sheep dips and others, e.g. pyrethroids. The Government, manufacturers, and the farming community need to work together to elucidate the problem as clearly as possible. There may also be long‐term, significant problems with financial compensation for afflicted individuals.

Abstract

Purpose

People with Down's syndrome are at increased risk of developing early onset Alzheimer's disease. It has been recommended that all adults with Down's syndrome receive baseline neuropsychological testing for dementia. In certain areas prospective screening of people with Down's syndrome takes place to ensure the early diagnosis of the condition. However, little has been published on the value of this type of screening. The purpose of this paper is to report on a prospective screening programme and asks whether the programme is effective in identifying dementia-related changes in people with Down's syndrome and whether the current screening intervals are appropriate.

Design/methodology/approach

All adults with Down's syndrome in Plymouth (UK) are identified and offered a comprehensive test battery at baseline at the age of 20 and then have testing biennially from 40 to 50 and annually after 50. All individuals diagnosed with dementia between 2001 and 2013 were identified and their case notes examined. The symptoms at the time of diagnosis were identified and whether these symptoms had been identified through the screening programme or by other routes were recorded. Prevalence data and age at diagnosis were also recorded.

Findings

In total, 26 people were diagnosed with dementia during the study period. Of these, the diagnosis of dementia followed concerns being identified during the routine screening programme in 54 per cent of cases. In the younger age group (age 40-49) 63 per cent of people were identified through the screening programme. At the time of diagnosis a mean of 5.5 areas of concern were in evidence.

Research limitations/implications

Limited by small sample size, however service development not original research. Implications for the use of prospective dementia screening in people with Down's syndrome.

Practical implications

Implications for how services choose to run their Down's syndrome and dementia screening programmes, including the frequency of screening.

Originality/value

The paper adds to a growing evidence base around the value of prospective dementia screening in people with Down's syndrome. It is also one of a few studies exploring the frequency of screening. Additionally, it adds further data about prevalence of dementia in people with Down's syndrome.

Purpose

Prediabetes can affect the central nervous system and lead to changes including impaired cognition and spatial memory. Isoflavones, especially genistein and daidzein, can increase spatial memory. Tempeh is a soy product with high levels of genistein and daidzein. The purpose of this paper is to determine the effect of tempeh on spatial memory of prediabetic Wistar rat.

Design/methodology/approach

A pre-and post-test randomised control group design was used. In total, 15 male Wistar rats were divided into three equal groups, Treatment 1 (T1), Treatment 2 (T2) and Treatment 3 (T3). All rats were injected with alloxan (120 mg/Kg body weight) to induce prediabetes. T1 was maintained on a diet of standard food for 14 days. T2 and T3 were given 9 g/200 g body weight and 18 g/200 g body weight of tempeh, respectively, for 14 days. The Morris water maze (MWM) test was used to test spatial memory.

Findings

While there were no statistically significant differences between the treatment groups, there was a difference with the tempeh-treated rats improving their spatial memory following tempeh ingestion with a decrease in the travel time to negotiate the MWM [T1(−8.36), T2 (−5.48) and T3 (3.66)]. Differences of travel time of the three groups was not significant (p = 0.677).

Originality/value

The result of the study indicates that tempeh may act as a functional food in reducing cognitive impairment associated with prediabetes and diabetes. However, as the result is not statistically significant, further research using animal models and human studies is warranted.

Abstract

Purpose

This paper aims to present a novel and cost-effective optical biosensor design by simple preparation method for detection of “parathion-methyl,” which is a model pesticide pose to public health and the environment.

Design/methodology/approach

The optical enzyme biosensor (TCA) for detection of pesticide “parathion-methyl” was developed on the basis of immobilization of tyrosinase enzyme on chitosan film by adsorption technique. The analytic performance of TCA was investigated by measuring its activity with Ultraviolet (UV) visible spectrophotometer.

Findings

Uniform porous network structure and protonated groups of chitosan film provided a microenvironment for tyrosinase immobilization evident from Fourier transform infrared (FTIR) spectroscopy and Atomic Force Microscopy analysis. TCA has a wide linear detection range (0-1.03 µM) with high correlation coefficient and it can detect the parathion-methyl concentration as low as 159 nM by noncompetitive inhibition kinetics. Using the TCA sensor both for ten times and at least 45 days without a significant loss in its activity are the indicators of its good operational and storage stability. Moreover, TCA can be applicable to tap water, providing a promising tool for pesticides detection.

Originality/value

This is the first time to use the in situ analytical technique that can improve the performance of optical enzyme sensor provided to control the pesticide residue better with respect to traditional techniques. The effect of organic solvents on the performance of optical enzyme biosensor was investigated. Inhibition kinetic of the solvents rarely encountered in literature was also studied besides the pH and temperature tolerance of the optical biosensor.

Purpose

The management of individuals with autism spectrum disorders (ASDs) requires a multimodal approach of behavioural, educational and pharmacological treatments. At present, there are no available drugs to treat the core symptoms of ASDs and therefore a wide range of psychotropic medications are used in the management of problems behaviours, co-occurring psychiatric disorders and other associated features. The purpose of this paper is to map the literature on pharmacological treatment in persons with ASD in order to identify those most commonly used, choice criteria, and safety.

Design/methodology/approach

A systematic mapping of the recent literature was undertaken on the basis of the following questions: What are the most frequently used psychoactive compounds in ASD? What are the criteria guiding the choice of a specific compound? How effective and safe is every psychoactive drug used in ASD? The literature search was conducted through search engines available on Medline, Medmatrix, NHS Evidence, Web of Science and the Cochrane Library.

Findings

Many psychotropic medications have been studied in ASDs, but few have strong evidence to support their use. Most commonly prescribed medications, in order of frequency, are antipsychotics, antidepressants, anticonvulsants and stimulants, many of them without definitive studies guiding their usage. Recent animal studies can be useful models for understanding the common pathogenic pathways leading to ASDs, and have the potential to offer new biologically focused treatment options.

Originality/value

This is a practice review paper applying recent evidence from the literature.
