Abstract

Purpose

The purpose of this paper is to carry out phytochemical investigations of different extracts of Eucalyptus globulus bark such as aqueous, methanolic and supercritical carbon dioxide fluid extract (SCFE) with ethyl acetate as entrainer. Three fractions (Eu 8, 9 and 10) containing steroidal δ‐lactone were isolated from SCF extract and the structure of Eu‐10 was earlier determined on the basis of NMR, HPLC‐MS, X‐Ray crystallography.

Design/methodology/approach

Column chromatography led to the isolation of flavonoids, tannins, steroids, etc. in different solvent systems. Isolated steroidal lactone (Eu‐8,9&10) of Withanolide series were tested for the presence of total phenolic content, total flavonoid content and the results were expressed as gGAE/100 g (TPC), and gQE/100 g (TFC), respectively. The antioxidant capacity was evaluated based on their ability to scavenge free radicals generated from ABTS, DPPH, FRAP and H2O2 by spectrophotometric method.

Findings

The result of the present study showed that different extracts of E. globulus bark and the isolated fractions, exhibited different antioxidant activity. This was due to the fact that they contained different amounts of flavonoid and phenolic compounds as per their ability to solubilize these compounds; the high scavenging property of E. globulus may be attributed to hydroxyl groups existing in the phenolic compounds. All the samples exhibited different extent of antioxidant activity (AOA) and showed higher potency when compared with BHT in scavenging action of DPPH free radical. Comparative data analysis showed SCF extract to be better than methanolic and aqueous extracts, both in terms of yield and AOA, while Eu‐10 was the best amongst purified fractions.

Practical implications

The present research has serious implications on identification of natural antioxidants from E. globulus. Natural antioxidants with better structure‐activity relationship are under investigation. Isolation of withanolide from Eucalyptus bark has opened newer horizon for its use.

Social implications

Collection of Eucalyptus bark from the forest (a forest waste) by women folk can be a source of revenue generation and thus has social implication as well. It is an important agro product.

Originality/value

The steroidal lactone (Eu‐10) showed highest radical scavenging effect even at IC50, thus the isolated lactone proved to be the best potential scavenger of free radicals amongst all crude extracts and the isolated fractions.

Abstract

Purpose

Flowers and fruits of Madhuca longifolia (Koenig) (mahua) tree are edible and used as traditional Indian medicines. The physicochemical properties of different parts of mahua are investigated. This study aims to estimate the different mineral contents, polyphenols compounds and antioxidant activities by 2,2-diphenyl-1-picrylhydrazyl inhibition, reducing power, free radical scavenging activity using 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) and ferric reducing antioxidant power assays of mahua flower, ripe and unripe fruit.

Design/methodology/approach

Flavonoids were identified and quantified in yellow flowers and fruits of M. longifolia tree by high-performance liquid chromatography with diode array detector. Low molecular weight carbohydrates were determined by the ICBio scan, a specific method for determining of carbohydrates. Mineral content is determined by laser-induced breakdown spectroscopy (LIBS) and atomic absorption spectroscopy. Physicochemical, nutritional and mineral properties of mahua flower, ripe and unripe fruit were investigated by the statistical approach of principal component analysis (PCA).

Findings

Ascorbic acid, gallic acid (GA), quercetin and myrcetin were the phenolic compounds identified and quantified in mahua flower and fruit extracts. Sugar profiling of mahua flowers and fruits confirmed the presence of inositol, sorbitol, mannitol, dextrose, fructose, sucrose, raffinose and maltose. The mineral content of Na, K, Mg and Ca was present in quite a good amount in all samples. Total phenolic content (TPC) was significantly high in mahua flower (25.3 ± 1.0 mg GA equivalent/g FW) followed by mahua unripe (15.8 ± 1.0 mg GA equivalent/g FW) and ripe fruit (14.3 ± 1.0 mg GA equivalent/g FW) at p = 5%. In contrast, total flavonoid contents (TFCs) were highest in ripe fruit, then mahua flower and unripe fruit. Positive correlations were predicted by PCA for mahua flower with TPC, antioxidant activity assays and minerals except for Na; ripe fruit with TFC and Na; and unripe fruit with maltose and sorbitol.

Originality/value

This study demonstrates the application of LIBS for the determination of elements present in the mahua flowers and fruits and reveals that mahua can be a good source of nutrients. Sugar profiling of mahua flower showed that it is a rich source of reducing and non-reducing sugar, proving that mahua flower juice can be used as a natural sweetener in the development of different food products, namely, biscuits, cookies, cake, jam, jelly, juice and squash.

Abstract

Purpose

The purpose of this paper is to study the structure‐activity correlation of four medicinal plants – Ocimum basilicum L., Piper betle L., Grewia asiatica L., Lantana camara L. in crude methanolic extract. These plants have been used in Ayurvedic preparation as alternative medicine. The contents of phenolic compounds (flavonoids, anthocyanin) and antioxidant capacities were evaluated.

Design/methodology/approach

Folin‐Ciocalteu method was used for the analysis of the phenolic compounds. Total flavonoids were also evaluated. DPPH, ABTS+, FRAP and H2O2 assays were used to evaluate antioxidant activity. Most of the assays were determined spectrometrically.

Findings

O. basilicum L. had the highest of total phenolics content (305.11 mg GAE/g), highest content of anthocyanins (15.427 mg/Kg) and total flavonoids (9.692 QE mg/g). The DPPH, ABTS+, FRAP and H2O2 assay indicated that these plants possessed considerable antioxidant activities.

Originality/value

The results also showed that O. basilicum and L. camara from red colored cultivars possessed high contents of phenolic compounds. FRAP assay showed AOA in the following sequence – Lantana camara>Ocimum basilicum>Piper betle>Grewia asiatica. Since L. camara showed highest antioxidant capacity thus it can be a potential resource for commercial antioxidant.

Abstract

Purpose

The purpose of this study is to evaluate the content of bioactive pigments in coloured callus of Azadirachta indica and to understand the correlation between the callus colours with their bioactive constituents, antioxidant properties and cytotoxicity. These assessments will yield valuable insight into the use of in vitro-derived pigments for possible use as functional natural colourants.

Design/methodology/approach

In this study, the authors have successfully developed a protocol to produce leaf-derived callus of various colours with enhanced content of bioactive pigments in A. indica through plant tissue culture. Comparative analysis of the pigments content (chlorophyll, carotenoid, phenolics and anthocyanins) in the coloured callus was conducted, followed by evaluation of its bioactive properties. The antioxidant properties against 2,2-diphenyl-1-picrylhydrazyl and 2,2′-azinobis(3-ethylbenzothiazoline-6-sulfonic acid) radicals, ferric reducing antioxidant power and cytotox activities of the coloured callus extracts were also reported.

Findings

Callus of various colours were successfully produced in A. indica through plant tissue culture, and their valuable pigment content and bioactivity were evaluated. The green callus contained the highest amount of anthocyanin, followed by brown and cream callus. The total anthocyanin contents in both the green and brown callus was more than two-fold higher than that in cream callus. Contrasting observation was obtained for total phenolic content (TPC), where the TPC of cream callus was significantly higher than that in brown callus. Nevertheless, the green callus also exhibited the highest TPC. Green callus also contained the highest amount of total chlorophyll and carotenoid, as well as exhibited the highest antioxidant potential, and was found to be the only extract with active cytotox activity against SKOV-3 cells. Correlation analysis revealed that the excellent bioactivity exhibited by the coloured extracts was strongly correlated with the bioactive pigments present in the callus.

Research limitations/implications

The major bioactive compounds identified in the methanolic extracts of A. indica coloured callus are anthocyanins, phenolics, chlorophylls and carotenoids. Future research work should include improvements in the extraction and identification methods, which may lead to detection and determination of other compounds that could attribute to its bioactivity, to complement the findings of the current study.

Practical implications

This analysis provides valuable information on the application of plant tissue culture as an alternative source for sustainable production of major pigments with medicinal benefits in A. indica for possible use as functional natural colourants.

Originality/value

A comparative study on bioactive pigment production in coloured callus from A. indica leaves and its antioxidant potential and cytotoxicity is original. To the best of the authors’ knowledge, this is the first report detailing a comparative evaluation on the production of coloured callus in A. indica and its relative biochemical composition and bioactive properties.

Abstract

Purpose

The purpose of this paper is to determine the polyphenol content, antioxidative potential and antiproliferative activity of Swiss chard from Montenegro, grown under different irrigation and fertilization regimes.

Design/methodology/approach

Swiss chard was grown in the open field (Lješkopolje, Montenegro) where it was subjected to different fertilization and irrigation regimes. Chard samples were analyzed for previously identified polyphenols and for antioxidant parameters. Additionally, in order to complete the biological activities, chard extracts were tested for antiproliferative activity against MFC-7 and HT-29 tumor cell lines.

Findings

The polyphenols identified in Swiss chard were flavonoids: vitexin-2?-O-xyloside, vitexin-6?-O-acetyl-2?-O-xyloside, vitexin-6?-O-malonyl-2?-O-xyloside and isorhamnetin-3,7-diglucoside. In the antioxidant tests, DPPH (2,2-diphenyl-1-picrylhydrazyl), and ABTS (2,2'-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid)), chard extract had values of 7.00 and 8.50 (mean values) µmol Trolox (6-hydroxy-2,5,7,8-tetramethylchroman-2-carboxylic acid) equ/d.w., respectively. The tested chard extracts inhibited cell proliferation at different concentrations (3.125–50.0 µg/mL) against the MCF-7 cell line, after 24 and 48 h of incubation. The antiproliferative activity, expressed in terms of IC50, was 32.97 and 86.45 µg/mL after 24 h of incubation and 20.76 and 23.33 µg/mL after 48 h of incubation, for treated and untreated chard extracts, respectively.

Originality/value

These data suggest Montenegrin Swiss chard grown under different irrigation and fertilization treatments can be considered as a functional food and should be included in an everyday diet. The collected data could help in the growth improvement of chard with functional food properties.

Purpose

The purpose of this paper is to evaluate the effect of different times and freezing temperatures on the antioxidant activity of raw human milk (HM) and the impact of light by different packaging on retinol level and the antioxidant activity of pasteurized HM.

Design/methodology/approach

Donor milks were homogenized to form the pool of the experimental study characterized by the evaluation of the effects of time (0, 2, 4, 8 and 15 days) freezing temperatures (−3°C, −8°C and −18°C) and the interference of the type of packaging on the antioxidant activity and retinol levels of HM.

Findings

The existing studies do not reveal the real impact of HM storage conditions adopted by human milk banks (HMB) in Brazil on their compounds, mainly in relation to the effects of temperature and freezing time and the incidence of light on retinol levels and antioxidant activity. In view of the already documented importance of these compounds for the growth, development and health of children, it is extremely important to assess their stability according to the procedures adopted by the banks. It has been observed in this study that lower freezing temperatures (−18°C) further preserve the antioxidant activity. It was found that the amber and transparent vials wrapped with aluminum foil allowed for greater retinol stability of HM, with values of 2.501±0.757 µmol/L and 4.991±0.825 µmol/L, respectively. On the contrary, there was no significant influence on antioxidant activity.

Originality/value

It is suggested that HMB store milk at lower temperatures and use glass jars that block the passage of light.

Abstract

Purpose

The purpose of this study was to evaluate the effect of black sesame germination on its nutritional composition and antioxidant activity.

Design/methodology/approach

Germination occurred at constant temperature and humidity (35 °C and 95 ± 5%) for 72 h. The levels of moisture, protein, lipids, ash, total dietary fibre and fractions, carbohydrates, calories, phenolic compounds and antioxidant capacity were determined.

Findings

With germination, there was a significant increase in moisture and protein content, as well as a reduction in the other parameters assessed in terms of nutritional composition. When analysing bioactive compounds, no significant change was found in the content of phenolic compounds; however, there was a reduction in antioxidant capacity according to the three methods tested, which may be related to the characteristics of the grain itself, the conditions used for germination, or even the process of extracting phenolic compounds for analysis. Other conditions for the germination of black sesame using different times and temperatures should be evaluated in future studies.

Originality/value

This is an original research article, which has an industrial and health impact by transmitting highly relevant responses on nutrition and food. The novelty of this research is the fact of studying the germination of black sesame on the nutritional composition (moisture, protein, lipids, ash, total dietary fibre and fractions, carbohydrates and calories), levels of phenolic compounds and antioxidant capacity. These results can help improve the procedures adopted by the food industries and also in nutritional conduct, as the germination process leads to a change in the nutritional composition, especially with regard to protein synthesis, which is relevant, once it is an interesting protein option. However, it also brings us responses to reductions in important nutritional compounds.

Abstract

Purpose

This study aims to focus on the preparation and characterization of the silver nanowire (AgNWs), as well as their application as antimicrobial and antivirus activities either with incorporation on the waterborne coating formulation or on their own.

Design/methodology/approach

Prepared AgNWs are characterized by different analytical instruments, such as ultraviolet-visible spectroscope, scanning electron microscope and X-ray diffraction spectrometer. All the paint formulation's physical and mechanical qualities were tested using American Society for Testing and Materials, a worldwide standard test procedure. The biological activities of the prepared AgNWs and the waterborne coating based on AgNWs were investigated. And, their effects on pathogenic bacteria, antioxidants, antiviral activity and cytotoxicity were also investigated.

Findings

The obtained results of the physical and mechanical characteristics of the paint formulation demonstrated the formulations' greatest performance, as well as giving good scrub resistance and film durability. In the antimicrobial activity, the paint did not have any activity against bacterial pathogen, whereas the AgNWs and AgNWs with paint have similar activity against bacterial pathogen with inhibition zone range from 10 to 14 mm. The development of antioxidant and cytotoxicity activity of the paint incorporated with AgNWs were also observed. The cytopathic effects of herpes simplex virus type 1 (HSV-1) were reduced in all three investigated modes of action when compared to the positive control group (HSV-1-infected cells), suggesting that these compounds have promising antiviral activity against a wide range of viruses, including DNA and RNA viruses.

Originality/value

The new waterborne coating based on nanoparticles has the potential to be promising in the manufacturing and development of paints, allowing them to function to prevent the spread of microbial infection, which is exactly what the world requires at this time.

Abstract

Purpose

This study aims to develop a green, economical and efficient ultrasonic-/microwave assisted extraction (UMAE) process for the extraction of anthocyanins.

Design/methodology/approach

After optimizing the extraction conditions by response surface methodology, three assays including DPPH, ABTS·+, FRAP were applied to analyze the antioxidant activity of the extracted anthocyanins. The stability under different temperatures, reductant concentrations and pHs was also discussed. The components of anthocyanins in blueberry were analyzed by HPLC-QTOF-MS2.

Findings

The optimal extraction parameters were ultrasonic power of 300 W, microwave power of 365.28 W and solid–liquid ratio of 30 (g/mL). The possible structures can be speculated as Delphinidin-3-O-galactoside, Delphinidin, Petunidin, Delphinidin-3-O-glucoside, Petunidin-3-O-glucoside, Cyanidin-3-O-glucoside. The results demonstrated that the UMAE can improve the yield of anthocyanins in shorter extraction time with higher activity.

Originality/value

The present study may provide a promising and feasible route for extracting anthocyanins from blueberries and studying their physicochemical properties, ultimately promoting the utilization of blueberry anthocyanins.

Abstract

Purpose

The purpose of this study was to evaluate beet and orange mixed juices added with lyophilized Lactobacillus acidophilus probiotic culture.

Design/methodology/approach

Two formulations of probiotic beet and orange mixed juices (1:1 e 1:2 v/v) were prepared, stored at 4°C for 28 days and were evaluated for probiotic survival, physicochemical stability, antioxidant activity and sensory acceptance. The probiotic culture was added in a lyophilized form and was not propagated in culture media or juice.

Findings

Physicochemical characteristics (pH, titratable acidity, total soluble solids and color) and antioxidant activity showed no undesirable alterations during storage. The number of probiotics followed established minimum requirement for probiotic foods (109 CFU/200mL) during the 28 days of storage. The juices showed good sensory acceptance and purchase intention, especially regarding the color of the product.

Practical implications

The beet and orange mixed juices are suitable carriers for L. acidophilus with a shelf life of at least 28 days at 4°C.

Originality/value

Direct addition of the probiotic culture could result in high probiotic survival in mixed juices, dispensing the propagation step.
